This video demonstrates and compares the various techniques of donor dissection in Descemet membrane endothelial keratoplasty (DMEK). In an experimental, nonrandomized, comparative study, 40 human sclerocorneal rims were divided into 4 groups, 10 in each. Four different techniques were tried by a single surgeon. The techniques were (1) reverse big bubble, (2) blunt knife dissection, (3) trephination-scoring and peeling, and (4) trephination and descemetorrhexis. Success of complete separation and ease of dissection and reproducibility were most with technique 4 by trephination and descemetorrhexis.
